Reconstitution of Respiratory Chain Enzyme Systems

ثبت نشده
چکیده

The reconstitution of a respiratory chain1 may be defined as the reassembly of the components to form an integral multienzyme system identical with the original. In the case of the succinate oxidase system, for example, the reconstituted particle shows all the properties of the original preparation as described in some of the papers of this series (e.g.(2-4)). This paper describes recent evidence which further reinforces the concept (cj. “Discussion” and “Conclusion”) of reconstitution. Preliminary reports (5-9) have already appeared on some facets of this communication.

Structural and Activity Comparison of Native, Apo and Reconstituted Tyrosinase

Background: Mushroom Tyrosinase a potent candidate in clinical studies known as polyphenol oxidase, is a metaloenzyme from the oxidase superfamily widely distributed from lower to higher life forms. It plays a crucial role in sclerotization of exoskeleton in insects, also responsible for skin pigmentation in mammalians. Objective: In this study, after reconstitution of MT by some metal io...

Long-term, high-dose aspirin therapy increases the specific activity of complex III of mitochondrial respiratory chain in the kidney of diabetic rats

Introduction: One of the main mechanisms by which diabetic complications occur is an alteration of the structure and function of proteins due to hyperglycemia. Aspirin (ASA) affects cellular pathways through different mechanisms, including glycation inhibition and antioxidant activity. The aim of the present study, as a follow up to our previous one, is to investigate the effect of long-term, h...

Reconstitution of Sue&ate-Coenzyme Q Reductase (Complex II) and Succinate Oxidase Activities by a Highly Purified, Reactivated Succinate Dehydrogenase*

Succinate dehydrogenase has been isolated in a highly purified form from succinate-coenzyme Q reductase preparations. The enzyme contains 1 mole of covalently bound flavin, 8 g atoms of iron, and 8 moles of acid-labile sulfide per 150,000 g of protein. It catalyzes succinate oxidation in the presence of phenazine methosulfate as electron acceptor at a rate of 26 to 32 pmoles per min X mg of pro...

Proline dehydrogenase from Escherichia coli K12. Reconstitution of a functional membrane association.

Soluble and membrane associated proline dehydrogenase differ in catalytic properties. The soluble enzyme transfers electrons from L-proline to exogenous electron acceptors. It has a high Km for L-proline (105 mM) and is insensitive to the respiratory chain inhibitors 5-ethyl-5-isopentyl-barbituric acid and cyanide. The membrane-associated enzyme transfers electrons from L-proline to O2 via the ...
